Interactions are made by either feeding a Pokémon a Berry or holding an Egg. When you interact with someone, they may interact back. This always helps when hatching Eggs or leveling up Pokémon.
You can either visit the Users Online list at the top of the page to find users to interact with, or use the "Open 10" or "Clickback" links to get ten random users or ten users that have interacted with you, respectively.
